Die beiden angehenden Rock-Stars Jack (JACK BLACK), genannt JB, und Kyle (KYLE GASS), alias KG stehen ziemlich auf dem Schlauch: Gerade erst haben sie die Gründung ihrer Rock-Band TENACIOUS D gefeiert, als KGs Eltern ihrem nicht mehr ganz so jungen Sprössling mitteilen, ihm künftig keine Unterstützung mehr zu zahlen. Doch was tun, wenn die Miete bereits überfällig, aber weder Kohle noch ein Band-Erfolg in Sicht sind?
Den beiden leidenschaftlichen Rock-Fans bleibt nur eine letzte Chance: der Sieg im Wettbewerb „Offenes Mikro“ in ihrer Stammkneipe Al’s Bar. Um den Wettbewerb zu gewinnen, müssten sie aber erst mal ein Meisterwerk schreiben. Das gestaltet sich schwieriger als erwartet…
Doch da kommt ihnen das Schicksal zu Hilfe: Sie entdecken, dass alle berühmten Rocker auf den Titelfotos ihrer gesammelten Rolling-Stone-Magazine eines gemeinsam haben: Sie scheinen alle das gleiche, mysteriöse Gitarren-Plektron zu benutzen. Als die beiden ihrem Verdacht nachspüren, bestätigt ihnen der Gitarrenhändler ihres Vertrauens (BEN STILLER) die schicksalhafte Verbindung zwischen dem Besitz des mysteriösen Plektrons und dem Aufstieg in den Rock’n’Roll-Olymp.
KB und JB versuchen fortan alles, um das Plektron des Schicksals zu ergattern – denn nur so werden sie vielleicht zur Größten Band der Welt.
